چکیده

Abstract Juvenile hormone (JH) is a major endocrine that mediates development, metamorphosis, and reproduction in insects. It binds directly to its methoprene-tolerant receptor recruits heterodimer partner form the JH–receptor complex then activates JH-inducible gene known as Krüppel homolog 1 (Kr-h1). There evidence this downstream factor mediating both physiological biochemical processes; however, functional mechanism of Kr-h1 largely unknown. Using economically important rice (Oryza sativa L.) pest Chilo suppressalis (Walker) (Lepidoptera: Crambidae) model, we used combination RNA interference (RNAi), high-throughput sequencing, real-time quantitative polymerase chain reaction (RT-qPCR) identify candidate transcription (TF) genes are regulated by Kr-h1. RNAi knockdown Krh1 identified Zinc finger proteins, ZBTB, THAP, PAX, MYB, HSF, Homeobox, HMG, CSD, basic helix-loop-helix, STAT, RHD, MBD families RT-qPCR confirmed levels these putative TFs indicated can induce or suppress expression proteins C. suppressalis. These results provide information required for in-depth research on other

Identification of genes differentially regulated by transcription factor, AP-2delta.

In an effort to better understand the transcriptional program regulated by transcription factor, AP-2delta, we used cDNA microarray to evaluate the relative expression of human genes in AD293 cells by exogenous expression of AP-2delta. Microarray showed 29 genes that were up-regulated and 39 genes with a down-regulated expression pattern. Among the identified genes were those encoding transcrip...

Identification of Putative Target Genes of the Transcription Factor RUNX2

Comparisons of the genomes of Neandertals and Denisovans with present-day human genomes have suggested that the gene RUNX2, which encodes a transcription factor, may have been positively selected during early human evolution. Here, we overexpress RUNX2 in ten human cell lines and identify genes that are directly or indirectly affected by RUNX2 expression. We find a number of genes not previousl...

Genome-wide identification of transcription factor target genes

Recent high-throughput techniques have allowed the identification of many DNA target sequences specifically bound by transcription factors at a genomic scale. UV or formaldehyde X-ChIP experiments performed on Drosophila embryos using Engrailed or Trithorax-like antibodies have led to the cloning and sequencing of in vivo Engrailed or Trl-recognized DNA fragments. The mining of such datasets ha...

Genome-wide identification of genes directly regulated by the pleiotropic transcription factor Spx in Bacillus subtilis

The transcriptional regulator Spx plays a key role in maintaining the redox homeostasis of Bacillus subtilis cells exposed to disulfide stress. Defects in Spx were previously shown to lead to differential expression of numerous genes but direct and indirect regulatory effects could not be distinguished. Here we identified 283 discrete chromosomal sites potentially bound by the Spx-RNA polymeras...

t-helper cell type-1 transcription factor t-bet is down-regulated in type 1 diabetes

t cells have been identified as key players in the pathogenesis of type 1 diabetes. however, the exact role of t-cell subpopulations in this pathway is presently unknown. the purpose of this study was to assess the expression pattern of two lineage-specifying transcription factors gata - 3 and t - bet , which are important in t helper type 1 (th1) and th2 cell development , respectively. gene e...
